Le génotypage complet d'un échantillon de queue de souris, y compris la digestion des tissus et la lecture de PCR, se fait en une heure et demie au moyen de Sigma SYBR Green Extract-N-Amp Tissue kit de PCR.

Abstract

Détection génomique de l'ADN par amplification par PCR et la détection sur un gel d'électrophorèse est une méthode standard que le génotype d'un échantillon de tissu est déterminé. Préparation classique des tissus pour la PCR-ADN souvent prêtes prendre plusieurs heures à plusieurs jours, selon l'échantillon de tissu. Le génotype de l'échantillon peut donc être retardée de plusieurs jours, ce qui n'est pas une option pour de nombreux types d'expériences. Ici nous démontrons le génotypage complète d'un échantillon de queue de souris, y compris la digestion des tissus et la lecture de PCR, en une heure et demie au moyen de Sigma SYBR Green Extract-N-Amp Tissue kit PCR. Premièrement, nous démontrons l'extraction de quinze minutes de l'ADN de l'échantillon de tissu. Ensuite, nous démontrons le temps réel de lecture de l'amplification par PCR de l'échantillon, ce qui permet l'identification d'un échantillon positif car il est amplifié. Ensemble, la lecture rapide de l'extraction en temps réel et permettent une identification rapide du génotype d'un type autre variété de tissus à travers la méthode fiable de la PCR.
