Lc-esi-ms/ms Quantification

LC-ESI-MS/MS quantification is an analytical method that measures specific compounds in complex samples by combining liquid chromatography with electrospray ionization and tandem mass spectrometry. Liquid chromatography first separates analytes, while electrospray converts them into gas-phase ions; the mass spectrometer then selects precursor ions, fragments them, and measures characteristic product ions, often against calibration standards or internal standards. In biochemistry, this approach supports sensitive, selective measurement of metabolites, peptides, proteins, lipids, and small-molecule drugs in biological specimens. Its quantitative data help characterize biochemical pathways, assess molecular changes, and evaluate disease biomarkers or therapeutic responses.

Visualization of miniSOG Tagged DNA Repair Proteins in Combination with Electron Spectroscopic Imaging (ESI)

Electron spectroscopic imaging can image and distinguish nucleic acid from protein at nanometer resolution. It can be combined with the miniSOG system, which is able to specifically label tagged proteins in transmission electron microscopy samples. We illustrate the use of these technologies using double-strand break repair foci as an example.

We present a protocol for non-targeted analysis using time of flight mass spectrometry as a perfect tool to identify pharmaceuticals in waters. We demonstrate the application of UV irradiation for their elimination. Analysis involving irradiation, compound isolation, identification and kinetic modelling of the degradation profiles is illustrated.

Here, we describe a chromatographic assay coupled with the ion mobility separation of peptide precursors followed by the high-resolution (~30,000) MS-detection of peptide fragments for the quantification of spiked peptide standards in a monoclonal antibody digest.
