Spot Intensity Histogram

A spot intensity histogram is a quantitative image-analysis tool that shows how signal intensities are distributed among detected spots or pixels within them. In fluorescence microscopy, software identifies spots against background and groups their measured intensities into bins, producing a distribution that can reveal signal strength, variability, and possible background contamination. In biochemistry, this analysis helps compare fluorescently labeled proteins, nucleic acids, or molecular complexes across samples and experimental conditions. Researchers can use histogram features to assess labeling consistency, distinguish weak from strong signals, evaluate detection thresholds, and support measurements of molecular abundance or localization in microscopy-based assays.

A probability histogram is a visual representation of a probability distribution. Similar a typical histogram, the probability histogram consists of contiguous (adjoining) boxes. It has both a horizontal axis and a vertical axis. The horizontal axis is labeled with what the data represents. The vertical axis is labeled with probability. Each rectangular bar in the histogram is 1 unit wide, which suggests that the area under each bar equals the probability, P(x), where x is 1, 2, 3, and so on.
