Anterior Stromal Keratectomy

Anterior stromal keratectomy is a corneal microsurgical technique that removes a controlled portion of the anterior stroma, the tissue layer beneath the epithelium, to treat or study superficial corneal abnormalities. During the procedure, a surgeon exposes the cornea and excises diseased, scarred, or structurally irregular stromal tissue while preserving deeper layers, allowing epithelial healing and stromal remodeling to occur. In bioengineering, this approach supports research on corneal wound repair, extracellular-matrix organization, biomaterial integration, and regenerative therapies. It can also provide a reproducible model for evaluating engineered tissues, cell-based treatments, and interventions designed to restore corneal transparency and mechanical function.

In this video, we demonstrate the culturing method of a cornea-bearing anterior eye segment from a porcine eye. This culture system serves as an experimental model to study eye injuries.

An In Vitro Flow Investigation of the Effect of Stromal Cells on Leukocyte Recruitment

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.
