Od 260/280

OD 260/280, also called the A260/A280 ratio, is a spectrophotometric measure used to assess nucleic acid concentration and purity in biological samples. The method compares absorbance at 260 nm, where DNA and RNA bases strongly absorb ultraviolet light, with absorbance at 280 nm, where aromatic amino acids in proteins absorb; changes in the ratio can therefore indicate protein or other contaminant carryover. Researchers use this measurement to evaluate DNA and RNA extracts before polymerase chain reaction, sequencing, cloning, and other molecular biology procedures. Interpreting the ratio alongside sample concentration and additional quality tests supports reliable experimental results.

The following protocol describes a novel method for chronic oral drug administration using an orally dissolving strip (ODS) in lieu of the more commonly used oral gavage method. We demonstrate that preclinical, oral drug delivery using the ODS method represents a safe, convenient, and humane alternative to oral gavage.
