Plant Tissue Extraction

Plant tissue extraction is the chemical process of isolating target compounds from plant materials for identification, analysis, or use in research and industry. It works by disrupting plant cells and transferring soluble constituents into a selected solvent, with extraction efficiency influenced by solvent polarity, temperature, particle size, contact time, and pH. Chemists apply techniques such as maceration, solvent extraction, and filtration to obtain pigments, lipids, proteins, nucleic acids, or secondary metabolites while minimizing unwanted components. The resulting extracts support phytochemical screening, natural-product discovery, quality control, environmental analysis, and the development of pharmaceuticals, foods, cosmetics, and agricultural products.

Source: Gulla, S., et al. Multi-locus Variable-number Tandem-repeat Analysis of the Fish-pathogenic Bacterium Yersinia ruckeri by Multiplex PCR and Capillary Electrophoresis. J. Vis. Exp. (2019).This protocol extracts genomic DNA from Yersinia ruckeri by suspending isolated colonies in ultrapure water and applying heat to lyse the cells. Centrifugation separates the DNA-containing supernatant from cell debris, yielding template DNA suitable for PCR-based genotyping and epidemiological analysis.

Plant Tissue Culture

2020

Plant tissue culture is widely used in both primary and applied science. Applications range from plant development studies to functional gene studies, crop improvement, commercial micropropagation, virus elimination, and conservation of rare species. Plant tissue culture depends on the ability of plant tissue to give rise to an entire new plant when provided with a growth medium and appropriate environment. This ability of plant cells or tissues is termed ‘totipotency.’ The fundamental steps...
