SDS-PAGE separation is an electrophoretic technique that resolves proteins by size, making it a fundamental method for analyzing complex protein mixtures in chemistry and biology. In the presence of sodium dodecyl sulfate (SDS), proteins unfold and acquire a nearly uniform negative charge-to-mass ratio; an applied electric field then drives them through a polyacrylamide gel, where smaller polypeptides migrate faster than larger ones. Researchers compare migration distances with molecular-weight standards to estimate protein size, assess sample purity, monitor purification, and evaluate changes in protein composition. The method also supports biochemical characterization and quality control by revealing distinct protein bands after staining.
