Plasma Concentration Fluctuations

Plasma concentration fluctuations are changes in the amount of a drug or biomolecule measured in blood plasma over time, an important consideration in clinical pharmacology and therapeutic monitoring. After administration, concentration rises during absorption and distribution, then falls as metabolism and excretion remove the substance; dose size, dosing interval, formulation, and patient physiology shape the resulting concentration-time profile. Understanding these fluctuations helps clinicians maintain concentrations within a therapeutic range, limit toxicity, and interpret pharmacokinetic measurements. The concept also supports dose adjustment, comparison of immediate- and extended-release formulations, and evaluation of treatment response.

Fluorescence Fluctuation Spectroscopy to Study Protein Homo-Oligomerization

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.
