Forskolin Swelling Assay

The forskolin swelling assay is a functional test that measures fluid-driven expansion of three-dimensional epithelial organoids after stimulation of cyclic AMP signaling, particularly to assess cystic fibrosis transmembrane conductance regulator (CFTR) activity. Forskolin activates adenylyl cyclase, increasing intracellular cyclic AMP and stimulating CFTR-mediated chloride and bicarbonate secretion; water follows osmotically, causing organoid swelling that can be quantified by imaging. In medicine, the assay helps characterize CFTR dysfunction in cystic fibrosis, evaluate patient-derived organoids, and measure responses to CFTR-modulating therapies. By linking molecular channel activity to a visible phenotype, it supports disease modeling, treatment selection, and preclinical drug development.
