Stereological Quantification

Stereological quantification is a set of statistical methods for estimating the three-dimensional number, volume, length, or surface area of biological structures from two-dimensional tissue sections, making complex anatomy measurable in medicine. It uses systematic random sampling and geometric sampling tools, such as point counting and unbiased counting frames, to reduce sampling bias and generate population-level estimates from representative sections. Applications include measuring cell populations, lesion volume, vascular networks, and tissue composition in histology, pathology, neuroscience, and experimental disease models. By providing reproducible quantitative outcomes, stereology supports comparisons between healthy and diseased tissues and strengthens the analysis of treatment effects.

The post-mortem assessment of myocardial infarction (MI) in rodents is based on quantification of the infarct on stained heart sections. We describe an accurate method to quantify the infarct size using systematic sampling of harvested rat hearts from base to apex and image analyses of trichrome-stained histological sections.
