Electron Microscopy Visualization

Electron microscopy visualization is an imaging approach that uses accelerated electrons to reveal ultrastructural details far smaller than those typically resolved by light microscopy. In transmission electron microscopy, electrons pass through ultrathin specimens, while scanning electron microscopy detects signals generated as a focused beam interacts with a specimen surface; electron scattering produces contrast that represents cellular architecture. In neuroscience, these methods visualize synapses, neuronal membranes, myelin, organelles, and other features of nervous tissue at nanometer-scale resolution. The resulting images support analysis of neural connectivity, cellular organization, and structural changes associated with development, injury, and neurological disease.

A method is described whereby quantum dot (QD) nanoparticles can be used for correlative immunocytochemical studies of epoxy embedded human pathology tissue. We employ commercial antibody fragment conjugated QDs that are visualized by widefield fluorescence light microscopy and transmission electron microscopy.
