Nerve Cord Dissection

Nerve cord dissection is a microsurgical technique used to isolate the central nerve cord from surrounding tissues for direct anatomical and physiological study in neuroscience. Under a stereomicroscope, researchers carefully remove external tissues while maintaining the cord’s structural integrity, allowing neural circuits, axons, and associated cells to remain accessible for observation or further analysis. The preparation supports investigations of nervous system organization, neuronal connectivity, development, and function, and can be combined with staining, imaging, electrophysiology, or molecular assays. By exposing neural tissue in a controlled preparation, nerve cord dissection helps link cellular structure with circuit activity and behavior.

To restore lost connectivity we utilize a peripheral nerve graft to provide a substratum for regenerating fibers in combination with neurotrophic factors and matrix-modulating enzymes to remove inhibitory molecules to promote long distance growth.

Whole Mount Preparation of the Adult Drosophila Ventral Nerve Cord for Giant Fiber Dye Injection

An in vivo dissection of the adult Drosophila ventral nerve cord (VNC) is demonstrated. This particular dissection method causes little damage to the VNC allowing the subsequent labeling of the giant fiber neurons with fluorescent dye for high resolution imaging.
