Fentanyl Analog Screening using LC-TIMS-TOF MS/MS

Le kit de dépistage analogique du fentanyl composé de 250 étalons analogues a été divisé en 14 groupes : 12 groupes de 17 analogues et 2 groupes de 16 analogues, afin d’éviter les interférences m/z. Chaque analogue est en outre caractérisé par son m/z, son temps de rétention (RT), sa mobilité (K) et son modèle de fragmentation MS/MS.
Des exemples de séparations isomériques sont illustrés à la figure 3 et à la figure 4 pour la formule chimique C22H28N2O2 et C21H28N2OS, respectivement. 
La trousse de dépistage analogue du fentanyl contient quatre isomères dont la formule chimique est le C22H28N2O2 (voir la figure 3). L’inspection de la figure 4 montre le chromatogramme ionique extrait [M+H]+ , les mobilogrammes, le motif isotopique et le modèle de fragmentation MS/MS. Le méthoxyacétyl fentanyl et le fentanyl carbamate se séparent bien dans les domaines LC et mobilité du para-méthoxyacétyl fentanyl et du B-hydroxyl fentanyl. Cependant, une séparation claire entre le para-méthoxyacétyl fentanyl et le B-hydroxyl fentanyl est obtenue par leurs modèles de fragmentation.
Le kit de dépistage analogique du fentanyl contient trois isomères dont la formule chimique est C21H28N2OS. L’inspection de la figure 4 montre le chromatogramme ionique extrait [M+H]+ , les mobilogrammes, le motif isotopique et le modèle de fragmentation MS/MS. L’a-méthyl thiofentanyl et le (±)-trans-3-méthyl thiofentanyl se séparent dans le domaine LC, mais pas dans le domaine de la mobilité. De plus, le (±)-trans-3-méthyl thiofentanyl et le (±)-cis-3-méthyl thiofentanyl se séparent dans le domaine de la mobilité et non dans le domaine LC. Cela montre la complémentarité de la LC et la séparation de mobilité, améliorant l’identification des espèces isomères par l’orthogonalité des domaines. La figure 4D résume ce résultat fourni par LC et mobilité.

Le profilage des métabolites non ciblés par chromatographie liquide ultra performante couplée à la spectrométrie de masse (UPLC-MS) est une technique puissante pour étudier le métabolisme.  Cet article décrit un flux de travail typique utilisé pour le profilage non ciblé des métabolites du sérum, y compris l’organisation et la préparation des échantillons, l’acquisition de données, l’analyse des données, le contrôle de la qualité et l’identification des métabolites.
