Karyotyping

During mitosis, DNA condenses into visible chromosomes. At this stage, the chromosome number and structure of a cell can be observed. This complete set is called a karyotype.Specific stains, such as Giemsa, reveal distinctive banding patterns that look like miniature barcodes.These stained chromosomes are then assembled into a complete chromosome map, called a karyogram, and arranged based on size, centromere position, and banding pattern.In humans, chromosomes are ordered from the largest, chromosome 1, to the smallest, chromosome 22, followed by the 23rd pair, the sex chromosomes.This layout helps scientists detect chromosomal abnormalities, such as an extra copy of a chromosome.Staining can also highlight different features along a chromosome&#x27;s arms.For example, specific regions, called cytogenetic bands, are labeled based on their position from the centromere to the telomere.When counting outward from the centromere, the short arm is labeled p, which stands for petite. The longer arm is labeled q, which is the next letter in the alphabet after p.This numbering scheme helps scientists pinpoint gene locations and study how changes in chromosome structure link to diseases and disorders.

Chromosome staining helps scientists find genetic disorders by showing the number and physical features of chromosomes. These features appear after DNA condenses into chromosomes during mitosis. Cytogeneticists then stop mitosis and stain the chromosomes to study their banding pattern. The pattern reveals details such as chromosome number, shape, and type.

A chromosome description like this is called a karyotype. To create one, scientists take an image of the stained chromosomes and identify each chromosome in the picture. They cut out the chromosomes and arrange them in pairs by size. This ordered display is called a karyogram. In a human karyogram, the 22 autosomes are labeled 1 through 22, from largest pair to smallest pair. The sex chromosomes are labeled X or Y.

A karyogram can reveal missing pieces, extra pieces, or an entire extra chromosome. These changes can point to genetic disease. One well-known example is Down syndrome, which Marthe Gautier, Jérôme Lejeune, and Raymond Turpin identified in 1959 as having a third copy of chromosome 21. Down syndrome is also called trisomy 21. People with Down syndrome often have mild to severe intellectual disability and delayed growth, but the effects can vary widely.

Down syndrome can begin when chromosome 21 fails to separate properly during meiosis, the cell division that makes sperm or egg cells. This can produce a germ cell with 24 chromosomes instead of 23. When that cell joins with another parent’s cell during fertilization, the zygote ends up with 47 chromosomes. In a small percentage of cases, only part of chromosome 21 is extra, and that extra piece is usually attached to another chromosome.

Modern chromosome studies use stains that give more detail than the early dyes used in cytogenetics. The first studies used a lichen-derived dye, but it was replaced by more stable dyes such as Giemsa. Giemsa stains some DNA regions more strongly than others because of base composition and chromatin structure. The result is G-banding, a reproducible pattern that is the same for individuals of a species. Because this pattern is predictable, abnormalities are easier to spot, and different banding methods can help diagnose different chromosomal problems.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules.
