מבוא

פרוטוקול

הערה: תכנון ניסוי ואישור אתי התקבלו מוועדת הביקורת המוסדית של UCL (IRB). כל הניסויים בוצעו בהתאם לרגולציה של משרד הפנים והנחיות בעלי חיים (הליך מדעי) חוק 1986 עם חקיקה מתוקנת של ההוראה האירופית 2010/63/האיחוד האירופי (2013). ארנבים נבדקו על ידי מנתח וטרינרי בשם (NVS) מעת לעת ופעמיים ביום על ידי קצין טיפול ורווחה לבעלי חיים בשם (NACWO) (בהתאם להנחיות ולתקנות של משרד הפנים). הם לא הראו שום סימן של כאב עד שהם מורדמים.

1. הכנת שתל גיד מהונדס רקמה (ET)

  1. כדי לפברק את הידרוג'ל הקולגן, הוסיפו 4 מ"ל של קולגן מסוג עכברוש מסוג 1 תמיסת קולגן מונומרית (2.15 מ"ג/מ"ל ב-0.6% חומצה אצטית עם 0.2% w/v של חלבון כולל) ו-500 מיקרול של 10x בינוני מינימלי חיוני. לנטרל את זה על ידי titrating נגד 5 M ו 1 M נתרן הידרוקסיד ולהוסיף 500 μL של מדיום הנשר שונה של Dulbecco (DMEM).

2. ייצור השתל

  4. להרדים בעלי חיים עם מינון תוך שרירי (i.m.) של Hypnorm (0.3 מ"ג / מ"ל) ולהמית על ידי מתן מנת יתר של פנטוברביטון.

3. טכניקת טנורהאפי חדשנית מפותחת

הערה: התפרים (ראה טבלה של חומרים) אינם נספגים ועשויים סטריאואיסומר גבישי איזוטקטי של פוליפרופילן, שהוא פוליאולפין ליניארי סינתטי. התפרים המשולבים העיקריים היו מורכבים בעיקר מ-3-0 והתפרים ההיקפיים היו 6-0. אלה היו שני התפרים העיקריים ששימשו בכל הניסויים.

תוצאות
