Arginine Agarose

Arginine agarose is an affinity chromatography medium in which arginine ligands are immobilized on agarose beads to capture and separate proteins, making it useful for biochemical purification. As a sample passes through the column, proteins interact reversibly with the arginine side chain through electrostatic forces, hydrogen bonding, and hydrophobic interactions; changes in salt concentration, pH, or competing arginine then weaken these interactions and promote elution. This approach can help isolate proteins from complex mixtures under relatively mild conditions, supporting studies of protein structure, function, purity, and interactions while providing an alternative to conventional ion-exchange or hydrophobic-interaction methods.

DNA Agarose Gel Electrophoresis

2021

Agarose gel electrophoresis is a laboratory technique commonly used to separate DNA fragments by size. However, it can also be used to isolate and purify DNA fragments using a gel extraction protocol. Gel extraction follows five major steps: running gel electrophoresis to separate fragments, isolating the individual bands, extracting DNA from those bands, and removing the dye and salts from the extracted mixture to obtain pure DNA. In cloning experiments, both the insert and vector DNA...
