Immunoglobulin G Fc

Immunoglobulin G Fc is the constant region of an IgG antibody that links antigen recognition to immune effector functions. Formed by paired heavy-chain CH2 and CH3 domains, the Fc region interacts with Fc gamma receptors on immune cells and can activate complement, while its N-linked glycan helps regulate receptor binding and inflammatory activity. In biochemistry, Fc structure and interactions are analyzed to understand antibody stability, cellular signaling, and immune clearance. Fc-containing fusion proteins, antibody therapeutics, and affinity-based assays use these properties to extend molecular half-life, recruit immune mechanisms, or enable selective purification and detection.

A Paper-Based Immunoassay for the Detection of Immunoglobulin G

2025

The video demonstrates a paper-based immunoassay, involving amine-functionalized cellulose discs cross-linked with glutaraldehyde. Covalent immobilization of IgG-Fc-specific capture antibodies, followed by the introduction of immunoglobulin G and horseradish peroxidase-conjugated detection antibodies, results in a color change confirming the presence of immunoglobulin G.

A Microfluidic Cartridge-Based Detection System for Allergen-Specific Immunoglobulin E

2025

This video demonstrates a protocol for detecting serum-specific immunoglobulin E using a microfluidic cartridge-based chemiluminescence system. Serum IgEs interact with distinct allergens immobilized in the reaction zone of the cartridge, which are subsequently detected with peroxidase-conjugated anti-IgE antibodies and a chemiluminescent substrate, allowing simultaneous detection of multiple allergen-specific IgEs.

Measuring the Activation of Fc-Mediated Effector Functions by HA Antibodies

2025

This video demonstrates an assay measuring the antibody-mediated activation of T cell effector functions. Adding an influenza virus hemagglutinin (HA)-specific monoclonal antibody to transfected mammalian cells expressing HA results in the formation of immune complexes with HA. Upon introducing engineered T cells expressing Fc receptors and a luciferase reporter, the T cells are activated by the antibody-HA complexes, and this activation is detected by adding a luciferase substrate and...

Measuring Immunoglobulin G Levels Against a Test Vaccine in a Mouse Serum

Compared with traditional affinity chromatography using protein A agarose bead-packed columns, protein A membrane adsorbers can significantly speed laboratory-scale isolation of antibodies and other Fc fragment-expressing proteins. Appropriate analysis and quantification methods can further accelerate protein processing, allowing isolation/characterization to be completed in one workday, instead of 20+ work hours.
