Antigen Uptake

Antigen uptake is the cellular process by which immune cells internalize foreign molecules or particles, enabling the immune system to detect and respond to potential threats. In antigen-presenting cells such as dendritic cells and macrophages, uptake occurs through mechanisms including endocytosis and phagocytosis, after which intracellular processing generates peptide fragments for presentation on major histocompatibility complex molecules. In bioengineering, researchers design nanoparticles, biomaterials, and vaccine-delivery systems to control antigen transport, cellular internalization, and immune activation. Understanding these steps supports the development of more effective vaccines, targeted immunotherapies, and engineered materials that regulate immune responses.

Radiolabeled Amino Acid Uptake Assay to Quantify Cellular Uptake of Amino Acids

2025

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

Development of an Antigen-driven Colitis Model to Study Presentation of Antigens by Antigen Presenting Cells to T Cells

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

Mitochondrial Calcium Uptake Assay: A Plate Reader-Based Method for Measuring Calcium Uptake in Isolated Mitochondria

2025

This video describes a plate reader-based assay to measure mitochondrial calcium uptake in isolated mitochondria. The assay analyzes the fluorescence signal of a calcium-sensitive fluorescent dye to study the kinetics of mitochondrial calcium uptake and calcium overload.

Viral Antigen Microarray Assay to Detect Antibody Isotypes in Serum Against Viral Antigen

In this video, we demonstrate the process for detecting antibody isotypes in multiple human serum samples using a microarray slide printed with antigenic subtypes of influenza virus strains.

Competence-Induced DNA Uptake in Staphylococcus aureus

Source: Cafini, F., et.al. Methodology for the Study of Horizontal Gene Transfer in Staphylococcus aureus. J. Vis. Exp. (2017)This video demonstrates competence-induced DNA uptake in Staphylococcus aureus through natural transformation. A chloramphenicol-sensitive strain engineered to constitutively express a sigma factor is cultured under nutrient-limiting conditions to enhance competence, then incubated with donor DNA carrying a resistance gene. Successful gene transfer is confirmed by the...
