Cytof Tuning

CyTOF tuning is the instrument-optimization process used to prepare a cytometry by time-of-flight mass spectrometer for accurate, sensitive single-cell measurements. During tuning, a metal-containing calibration solution is introduced and the instrument adjusts plasma conditions, ion optics, detector response, and mass calibration to maximize signal stability while minimizing background and oxide formation. Reliable tuning supports consistent detection of metal-tagged antibodies and other probes across complex samples, making it essential for immunophenotyping, biomarker analysis, and bioengineering studies of cell states, engineered tissues, and therapeutic responses. Proper performance checks also improve data comparability between experiments and help identify instrument problems before sample acquisition.
