Transcellular Interactions

Transcellular interactions are functional exchanges between distinct cells that allow one cell to influence another through direct contact, signaling, or transfer of biological materials. They occur when membrane-bound ligands engage receptors, adhesion molecules stabilize cell-cell contacts, or structures such as gap junctions and extracellular vesicles move signals and molecules across cellular boundaries. In biological techniques, researchers study these interactions using co-culture systems, cell-contact assays, and engineered tissue or barrier models. Such approaches clarify how cells coordinate immune responses, development, tissue repair, and disease progression, while supporting drug testing and the design of more physiologically relevant experimental models.

Transcellular Transport of Solutes

2023

Transcellular transport of solutes is the movement of substances like monosaccharides and amino acids through polarized cells. This transport mechanism is primarily seen in epithelial and endothelial cells aided by membrane transport proteins such as channels and transporters. The tight junctions between these cells confine the membrane proteins to the two sides of the cell. The epithelial cells have distinct apical and basolateral domains. In contrast, the endothelial cells show the luminal...

Cell Aggregation Assay to Detect Trans Interactions between Cell Adhesion Molecules

In this video, we demonstrate the technique to measure transcellular interactions between synaptic cell adhesion molecules through the HEK cell aggregation assay. In this assay, an equal number of HEK cells expressing a desired presynaptic cell adhesion molecule and a fluorescent protein are mixed with HEK cells expressing a post-synaptic cell adhesion molecule and another fluorescent protein. The interaction between the cell adhesion molecules forces the cells to aggregate, which can be...

Mechanisms of Drug Absorption: Paracellular, Transcellular, and Vesicular Transport

Drugs need to permeate cell membranes to reach their target sites after administration. Orally administered drugs must transcend intestinal epithelial membrane barriers to infiltrate the systemic circulation. Drugs with a molecular weight of less than 500 Daltons diffuse through gaps between neighboring cells, called paracellular pathways. However, most drugs use the transcellular route, traversing directly through the cell membranes via two mechanisms: passive and active transport. Passive...

Time-Lapse Imaging to Monitor the Transcellular Spreading of Prion-Like Proteins in Transgenic Nematodes

This video demonstrates the procedure for in vivo imaging of C. elegans expressing prion-like proteins tagged with red fluorescent protein. The protocol involves immobilizing nematodes on an agarose pad for time-lapse confocal microscopy, revealing protein aggregation and transport within and between cells. This technique is crucial for studying protein aggregation related to neurodegenerative diseases.
