Ceramic Bead Homogenization

Ceramic bead homogenization is a mechanical sample-preparation technique that disrupts biological cells and tissues to release intracellular components for analysis. It works by vigorously agitating a sample with hard ceramic beads, producing repeated collisions, impact, and shear forces that break cell walls and membranes; buffer conditions help stabilize the released material and support efficient lysis. The method is useful for processing difficult samples, including microorganisms and tissue, before extracting DNA, RNA, proteins, or other biomolecules. Its speed, scalability, and compatibility with closed tubes make it valuable in molecular biology, microbiology, diagnostics, and research workflows requiring consistent sample disruption.

This protocol presents a technique for probing protein-protein interactions using glutathione-linked donor beads with GST-fused TPR-motif co-chaperones and acceptor beads coupled with an Hsp90-derived peptide. We have used this technique to screen small molecules to disrupt Hsp90-FKBP51 or Hsp90-FKBP52 interactions and identified potent and selective Hsp90-FKBP51 interaction inhibitors.
