Glass Bead Homogenization

Glass bead homogenization is a mechanical cell-disruption technique that uses small glass beads to break open biological samples and release intracellular contents for analysis. During vigorous shaking or mixing, collisions between the beads and suspended cells generate impact and shear forces that rupture cell envelopes, while buffer conditions and temperature help preserve the released molecules. The resulting lysate supports protein extraction, nucleic-acid analysis, enzyme assays, and microbiological studies, particularly for organisms or tissues that resist gentler methods. Because it processes samples efficiently without relying on chemical lysis, glass bead homogenization is valuable in routine biology and reproducible laboratory workflows.

This protocol presents a technique for probing protein-protein interactions using glutathione-linked donor beads with GST-fused TPR-motif co-chaperones and acceptor beads coupled with an Hsp90-derived peptide. We have used this technique to screen small molecules to disrupt Hsp90-FKBP51 or Hsp90-FKBP52 interactions and identified potent and selective Hsp90-FKBP51 interaction inhibitors.
