Kinetic Analysis

Kinetic analysis is the quantitative study of how fast biological processes occur and how their rates change with experimental conditions. It commonly tracks substrate depletion or product formation over time, then uses initial reaction rates to relate velocity to variables such as substrate concentration, enzyme amount, temperature, or inhibitors; in enzyme studies, Michaelis-Menten behavior can estimate catalytic parameters. In biology, these measurements help characterize enzyme function, compare catalytic efficiency, identify competitive or other inhibition patterns, and distinguish rate-limiting steps in metabolic or cellular pathways. By linking molecular interactions to measurable outcomes, kinetic analysis supports drug screening, pathway modeling, and interpretation of how mutations or environmental changes alter biological activity.

The Importance of Correct Protein Concentration for Kinetics and Affinity Determination in Structure-function Analysis

We apply label-free protein interaction analysis using Biacore X100 for structure-function analysis of the binding of several cystatin B mutants to papain through kinetic characterization. Calibration-free concentration analysis (CFCA) measures the concentration of protein with retained binding activity without the need for a standard curve. We show that confirmation of concentrations using CFCA increases the reliability of the kinetic analysis and that kinetic constants can reliably be...

Enzyme Assays and Kinetics

2023

Enzyme kinetics describes the catalytic effects of enzymes, which are biomolecules that facilitate chemical reactions necessary for living organisms. Enzymes act on molecules, referred to as substrates, to form products. Enzyme kinetic parameters are determined via assays that directly or indirectly measure changes in substrate or product concentration over time. This video will cover the basic principles of enzyme kinetics (including rate equations) and kinetic models. The concepts governing...

Kinetic Molecular Theory: Molecular Velocities, Temperature, and Kinetic Energy

2020

The kinetic molecular theory qualitatively explains the behaviors described by the various gas laws. The postulates of this theory may be applied in a more quantitative fashion to derive these individual laws. Collectively, the molecules in a sample of gas have average kinetic energy and average speed; but individually, they move at different speeds. Molecules frequently undergo elastic collisions in which the momentum is conserved. Since the colliding molecules are deflected off at different...
