Peritoneal Incision

Peritoneal incision is a deliberate surgical opening of the peritoneum, the thin serous membrane that lines the abdominal cavity and covers many abdominal organs. The procedure creates controlled access by separating or cutting this tissue while limiting damage to nearby structures, then allowing the incision to be closed so the membrane can heal and restore the abdominal barrier. In biology and biomedical research, peritoneal incisions support abdominal surgery, tissue sampling, implantation procedures, and experimental studies of wound healing, inflammation, adhesions, and tissue repair. Careful technique is essential because disruption of the peritoneum can influence recovery and postoperative tissue responses.

The peritoneal cavity in mammals contains different immune cell populations crucial for innate immune responses. An efficient isolation method is required for biochemical and functional analyses of these cells. Here we provide a comprehensive method for the isolation of peritoneal cavity cells in the mouse.

This video demonstrates a peritoneal lavage method for isolating mature connective tissue-type mast cells from the peritoneal cavity of adult mice. Upon isolating a mixture of primary immune cells from the peritoneal cavity, selective proliferation of mast cells is performed under appropriate culture conditions.
