Second Dimension Sds-page

Second-dimension SDS-PAGE is the electrophoretic step in two-dimensional gel electrophoresis that separates proteins by molecular mass after isoelectric focusing has separated them by isoelectric point. In this stage, proteins are treated with sodium dodecyl sulfate (SDS), which denatures them and gives them a similar negative charge-to-mass ratio, then migrate through a polyacrylamide gel toward the positive electrode, with smaller proteins moving faster. The resulting vertical separation complements the first dimension, producing a two-dimensional protein pattern that can resolve complex biological samples. Researchers use these maps to compare protein expression, identify disease- or treatment-associated changes, and select spots for downstream mass spectrometric analysis.
