Homogeneous Spheroid Size

Homogeneous spheroid size refers to the consistent diameter or volume of three-dimensional cell aggregates formed under controlled culture conditions. In cancer research, uniform spheroids develop when cells are distributed at standardized densities and maintained in methods such as low-adhesion or hanging-drop culture, producing comparable diffusion distances for oxygen, nutrients, and therapeutic compounds. Controlling size improves experimental reproducibility and helps researchers distinguish biological treatment effects from variations in aggregate geometry. Standardized spheroids are therefore useful for modeling tumor architecture, evaluating drug penetration and cytotoxicity, and studying how gradients of oxygen, nutrients, and metabolites influence cancer cell behavior.

We present a high-throughput image analysis software application to measure the size of three-dimensional tumor spheroids imaged with bright-field microscopy. This application provides a fast and effective way to examine the effects of therapeutic drugs on spheroids, which is beneficial for researchers who wish to use spheroids in drug screens.

Mechanical Mapping of Spheroids Using Brillouin Spectroscopy

This article describes a protocol for mechanical characterisation of living spheroids within a 3D matrix using Brillouin micro-spectroscopy. This all-optical method enables spheroid visualisation with microscale resolution and quantitative mechanical properties. This approach has implications for mechanical phenotyping; for instance, determining pathological states of tumor spheroids within a 3D microenvironment.
