Microbial Dna Isolation

Microbial DNA isolation is the process of extracting and purifying genetic material from bacteria, fungi, or other microorganisms for molecular analysis. The method typically disrupts microbial cells through chemical, enzymatic, or mechanical lysis, then removes proteins, cellular debris, and inhibitors to recover DNA suitable for amplification or sequencing. In cancer research, isolated microbial DNA helps characterize tumor-associated and tissue-resident microbial communities, investigate interactions between microbes and host cells, and identify microbial signatures linked to disease or treatment response. Reliable DNA isolation supports microbiome profiling, pathogen detection, biomarker development, and studies of how microbial composition may influence cancer biology.

This video describes DNA isolation protocol from stool samples of colon tumor-bearing mice. The extracted DNA is amplified using PCR, and the product is further cleaned using a magnetic beads-based approach. This method can be used to determine the effect of environmental enrichment on colon microbiota and animal mortality.

We provide an improved protocol for extracting high molecular weight DNA from hypersaline microbial mats. Microbial cells are separated from the mat matrix prior to DNA extraction and purification. This enhances the concentrations, quality, and size of the DNA. The protocol may be used for other refractory samples.

DNA isolation protocols can be fast and straightforward or complex and time-consuming depending on the type and quality of DNA required for further processing. For example, plasmid DNA extraction is a bit more complicated than genomic DNA extraction because of the need for an appropriate lysis method to separate plasmid DNA from gDNA during isolation. However, for specific applications, such as long-range DNA sequencing that require a good yield of high- quality DNA samples, we need to follow...
