Biological Sample Preparation

Biological sample preparation is the set of procedures used to convert tissues, cells, fluids, or other biological materials into suitable samples for chemical analysis. It typically involves collection, homogenization, extraction, filtration or centrifugation, and sometimes concentration or purification to isolate target compounds while removing interfering substances. Careful control of factors such as solvent choice, pH, temperature, and handling time helps preserve analyte stability and improve measurement accuracy. In chemistry, effective sample preparation supports chromatography, mass spectrometry, spectroscopy, and biochemical assays, enabling reliable identification and quantification of metabolites, proteins, drugs, contaminants, and other compounds in complex biological matrices.

Source: Peiman Shahbeigi-Roodposhti and Sina Shahbazmohamadi, Biomedical Engineering Department, University of Connecticut, Storrs, Connecticut A scanning electron microscope (SEM) is an instrument that uses an electron beam to nondestructively image and characterize conductive materials in a vacuum. As an analogy, an electron beam is to the SEM as light is to the optical microscope. The difference is that the electron microscope yields images of much higher resolution and magnification. The...

Chemical Drying: Biological Sample Preparation for Scanning Electron Microscopy (SEM)

Scanning electron microscopy (SEM) is a powerful tool to image—primarily—the surface features of a biological sample. Proper sample preparation, however, is critical to obtaining quality images. This video focuses on the dehydration and drying steps of preparing intact adult Drosophila for SEM.

X-ray Diffraction of Biological Samples

X-ray diffraction or XRD is an analytical tool that utilizes X-rays to study ordered structures such as crystalline organic and inorganic samples, polycrystalline materials, proteins, carbohydrates, and drugs. According to Bragg's law, when X-rays strike the sample positioned on a stage, the rays are scattered by the electron clouds around the sample atoms. The X-ray diffraction or scattering is caused by constructive interference of the X-ray waves that reflect off the internal crystal...
