Hydroxide Base Avoidance

Hydroxide base avoidance is a strategy in chemistry that replaces hydroxide ions with alternative bases when hydroxide could cause unwanted reactions or reduce selectivity. The approach relies on choosing a base that can remove a proton without introducing a strongly nucleophilic hydroxide species, helping limit hydrolysis, substitution, or other side reactions under the reaction conditions. Chemists use this principle in organic synthesis and other solution-based reactions to protect sensitive functional groups, improve product purity, and control reaction pathways. Selecting an appropriate nonhydroxide base requires considering basicity, nucleophilicity, solvent compatibility, temperature, and the stability of the starting materials and products.

This article describes a simple, low-cost method of recording the lawn avoidance behavior of Caenorhabditis elegans, using readily available items such as a smartphone and a light emitting diode (LED) light box. We also provide a Python script to process the video file into a format more amenable for counting.
