Pharmacokinetic Assumptions

Pharmacokinetic assumptions are simplifying principles used to predict how a drug is absorbed, distributed, metabolized, and excreted in the body, supporting clinical dosing decisions. Models may assume dose-proportional exposure, first-order elimination, consistent bioavailability, and stable clearance and volume of distribution; these assumptions connect drug dose to plasma concentration over time and help estimate half-life and steady-state levels. In clinical pharmacology, evaluating whether these conditions apply guides dose selection, therapeutic drug monitoring, and interpretation of concentration data. Recognizing departures caused by organ dysfunction, drug interactions, or nonlinear kinetics improves treatment safety and helps refine pharmacokinetic models.
