Embryo Anesthesia Technique

Embryo anesthesia technique is the controlled use of anesthetic agents to temporarily reduce movement and responsiveness in developing embryos, enabling safe observation and experimental manipulation in developmental biology. Embryos are exposed to an appropriate anesthetic solution under defined concentration and exposure-time conditions; the agent suppresses neuronal excitability and muscle activity, producing reversible immobilization while preserving viable development when properly applied. This technique supports microscopy, microinjection, surgical manipulation, phenotypic analysis, and time-lapse imaging by limiting motion-related artifacts and handling stress. Careful optimization of dose, developmental stage, and recovery conditions is essential for reliable results and accurate interpretation of developmental processes.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN The Guide for the Care and Use of Laboratory Animals ("The Guide") states that pain assessment and alleviation are integral components of the veterinary care of laboratory animals. The definition of anesthesia is the loss of feeling or sensation. It is a dynamic event involving changes in anesthetic depth with respect to an animal's metabolism, surgical stimulation, or variations in the...

The topical anesthetic lidocaine was atomized using a high oxygen flow through the working channel of a flexible intubating endoscope to achieve topical airway anesthesia for awake endotracheal intubation. We prefer this modified spray-as-you-go technique for endoscopic intubation to classical bolus application because of higher patient satisfaction and better compliance.

In this video, we describe the procedure for vitrification of harvested rabbit embryos at desired development stage using ethylene glycol and dimethyl sulfoxide as cryoprotectants and cryogenic straw as the carrier system. The vitrified embryos have high survival rate and can be stored for an extended period until use.
