Qrt-pcr Quantitation

qRT-PCR quantitation is a molecular method for measuring the abundance of specific RNA transcripts, enabling researchers to compare gene expression across samples and developmental stages. During the assay, reverse transcriptase converts RNA into complementary DNA, which is amplified with sequence-specific primers while fluorescence monitors product accumulation; cycle threshold values indicate the starting template amount. Researchers can use reference genes for relative expression analysis or standard curves for absolute quantitation. In developmental biology, qRT-PCR helps track tissue-specific and stage-specific gene regulation, assess responses to genetic or environmental perturbations, and connect transcriptional changes with processes such as differentiation, pattern formation, and organ development.

Here, we present a protocol for the development and validation of a quantitative PCR method used for the detection and quantification of EHV-2 DNA in equine respiratory fluids. The EHV-2 qRT-PCR validation protocol involves a three-part procedure: development, characterization of qRT-PCR assay alone, and characterization of the whole analytical method.
