Cas9 Mrna Preparation

Cas9 mRNA preparation is the process of generating purified messenger RNA that encodes the Cas9 genome-editing enzyme for use in genetic engineering. Typically, a DNA template containing the Cas9 coding sequence undergoes in vitro transcription, followed by incorporation or addition of a 5′ cap and poly(A) tail and purification to remove DNA, enzymes, and abnormal RNA products. After delivery into cells with a guide RNA, the mRNA is translated transiently into Cas9, which creates a targeted DNA break that enables gene disruption or sequence modification through cellular repair pathways. This approach supports CRISPR experiments, functional genomics, disease modeling, and potential therapeutic development while limiting prolonged Cas9 expression.
