Streptavidin Bead Binding

Streptavidin bead binding is a molecular capture method that uses the exceptionally strong interaction between streptavidin and biotin to isolate specific biomolecules. In this process, biotinylated DNA, RNA, proteins, or molecular probes bind to streptavidin-coated beads, allowing the target complex to be retained while unbound material is removed through washing; bound molecules can then be released or analyzed. In genetics, the method supports nucleic acid purification, affinity capture, sequencing workflows, and isolation of protein-DNA or protein-RNA complexes. Its high specificity and adaptable bead-based format improve target enrichment and simplify downstream genetic analysis.

Source: Pohl, M. O. & Stertz, S. Measuring Attachment and Internalization of Influenza A Virus in A549 Cells by Flow Cytometry. J. Vis. Exp. (2015)This video demonstrates the use of a streptavidin blocking assay to distinguish surface-bound from internalized influenza virus in human lung epithelial cells. Comparing fluorescence signals shows that a higher intensity after incubation indicates successful viral internalization.

A Multiplex Bead-Based Immunoassay to Quantify Multiple Cytokine Targets

2025

This video demonstrates a multiplex bead-based immunoassay for quantifying multiple cytokine targets in a sample. A mixture of distinct microspheres conjugated to cytokine-specific antibodies is used to bind the cytokines present in the sample. Streptavidin-tagged reporters are immobilized on the bound-cytokines using biotinylated detection antibodies. Flow cytometry identifies the bound cytokines based on the unique sizes and internal fluorescence intensities of the microspheres, while the...
