Mouse Tibia Isolation

Mouse tibia isolation is a laboratory technique for excising the tibia from a mouse while preserving bone and marrow for downstream analysis. The procedure typically involves exposing the lower limb, removing muscle and connective tissue, separating the tibia from adjacent joints, and processing the bone to recover marrow-derived cells or prepare bone tissue, with aseptic handling helping limit contamination. In immunology and infection research, isolated tibiae provide material for studying hematopoietic cell populations, inflammatory responses, pathogen-associated changes, and bone or marrow pathology, supporting flow cytometry, culture, molecular assays, and histological evaluation.

We describe a protocol for isolation and purification of neutrophils from mouse bone marrow by density gradient centrifugation and for neutrophil labeling using CellTracker dyes. This represents a simple, fast, reproducible and economical method for obtaining large numbers of neutrophils for downstream functional studies or adoptive transfer and tracking experiments.
