Sigma Factor Purification

Sigma factor purification is a biochemical method for isolating bacterial sigma factors, transcription components that direct RNA polymerase to particular promoter sequences and thereby shape gene expression during infection and stress. The process generally involves producing the target protein, disrupting bacterial cells, and separating it from other proteins by chromatography, often through affinity capture of a tagged sigma factor followed by polishing and purity assessment. Purified sigma factors enable reconstitution of transcription reactions, analysis of promoter recognition and regulatory switching, and characterization of bacterial adaptation pathways that influence host colonization, immune evasion, and antimicrobial susceptibility.

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study.
