Pre-embedding Immunostaining

Pre-embedding immunostaining is an antibody-based method in which fixed tissue is labeled before it is embedded for sectioning, allowing researchers to localize specific proteins within cells, axons, dendrites, or synapses. The process typically uses tissue permeabilization so primary antibodies can bind target antigens, followed by labeled secondary antibodies or enzyme-linked detection that generates a visible reaction product before embedding and microscopy. In neuroscience, the method connects molecular identity with cellular and subcellular structure, supporting studies of neurotransmitter systems, receptor distribution, and synaptic organization; combined with electron microscopy, it can reveal where labeled molecules occur in relation to neural ultrastructure.
