Pre-embedding Technique

The pre-embedding technique is a tissue-labeling method in which target molecules are visualized before a biological specimen is embedded and sectioned, preserving their anatomical context for microscopy. In neuroscience, fixed brain tissue is incubated with a primary antibody and a labeled secondary antibody, often followed by an enzymatic or electron-dense reaction product, before embedding in resin or another medium. The resulting label reveals the cellular or subcellular location of proteins, receptors, or synaptic components within tissue sections. This approach supports anatomical mapping and ultrastructural analysis, linking molecular identity with neuronal morphology, connectivity, and synaptic organization.

We describe a novel experimental technique that we call Minimally Invasive Muscle Embedding (MIME), which is based on the evidence that skeletal muscle tissue contains viable myogenic cells that can facilitate donor-cell-mediated myogenesis when implanted into a host muscle.

In eukaryotic cells, transcripts made by RNA polymerase are modified and processed before exiting the nucleus. Unprocessed RNA is called precursor mRNA or pre-mRNA to distinguish it from mature mRNA.
