competitivo e non competitivo.

Un inibitore competitivo

diminuisce il numero

di enzimi con substrato
disponibile a legarsi.

Al contrario, un inibitore non competitivo

si legherà lontano dal sito
attivo, pur influenzandolo,

cambiando la forma

e riducendo notevolmente l'affinità

per il legame del
substrato al sito attivo,

impedendo il corretto
funzionamento dell'enzima.

Enzyme Inhibition

Gli inibitori sono molecole che riducono l'attività degli enzimi legandosi all'enzima. In una cellula normalmente funzionante, gli enzimi sono regolati da una varietà di inibitori. Farmaci e altre tossine possono anche inibire gli enzimi. Alcuni inibitori si legano al sito attivo dell'enzima, mentre altri inibiscono l'attività enzimatica legandosi ad altri siti sulla struttura proteica.

Gli inibitori competitivi occupano il sito attivo di enzimi, rendendoli incapaci di ospitare il substrato. Tuttavia, concentrazioni sufficientemente elevate del substrato possono superare l'inibitore; di conseguenza, gli inibitori competitivi rallentano un tasso di reazione iniziale degli enzimi, ma non influiscono sul tasso massimo dell'enzima. Un esempio di inibitore competitivo è il farmaco "disulfiram", usato per trattare l'alcolismo cronico. Quando l'alcol viene ingerito, viene normalmente convertito in acetaldeide, che viene poi convertito in acetil coenzima A dall' acetaldeide deidrogenasi. Disulfiram si lega e occupa il sito attivo dell' acetaldeide deidrogenasi, rendendo l'enzima incapace di eseguire questa conversione. Di conseguenza, un paziente che assume disulfiram inizia immediatamente a sperimentare sintomi simili a postumi di una sbornia, come mal di testa, riducendo così il consumo di alcol.

Gli inibitori non competitivi si legano a siti distinti sull'enzima, lontano dal sito attivo. Questi sono chiamati siti alsicici e quando le molecole si legano ad essi, la forma del sito attivo viene modificata in modo tale che l'enzima ha una minore affinità per il substrato. Poiché gli inibitori non competitivi non occupano il sito attivo, la presenza di substrato aggiuntivo non è in grado di superare l'inibizione non competitiva e l'enzima non è in grado di raggiungere il suo tasso di reazione massimo.

Il legame covalente tra un inibitore e un enzima è solitamente irreversibile, come nel caso di alcune tossine. La maggior parte degli inibitori regolatori normalmente attivi nella cellula interagisce con gli enzimi da interazioni deboli. Questo tipo di legame è reversibile e utile per la regolazione dei processi metabolici. Enzyme inhibitors are molecules that reduce enzyme activity by binding to enzymes. Cells use these inhibitors to help regulate normal enzyme function. Drugs and toxins can also block enzymes and change how they work.

Some inhibitors bind at the active site, which is the part of the enzyme where the substrate normally fits. Competitive inhibitors do this by occupying the active site and preventing the substrate from binding. If the substrate concentration becomes high enough, it can outcompete the inhibitor. For that reason, competitive inhibitors slow the initial reaction rate but do not change the enzyme’s maximum rate.

A clear example is disulfiram, a drug used to treat chronic alcoholism. Alcohol is normally converted to acetaldehyde and then to acetyl coenzyme A by acetaldehyde dehydrogenase. Disulfiram binds to the active site of acetaldehyde dehydrogenase and blocks this conversion. This causes hangover-like symptoms, such as headache, and helps decrease alcohol consumption.

Noncompetitive inhibitors bind at a different part of the enzyme. These binding sites are called allosteric sites. When a noncompetitive inhibitor binds, it changes the shape of the active site and lowers the enzyme’s affinity for the substrate. Extra substrate cannot overcome this effect, so the enzyme cannot reach its maximum reaction rate.

Some inhibitors form covalent bonds with enzymes. This binding is usually irreversible, as with some toxins. Most inhibitors that regulate metabolism in cells bind more weakly. That weak binding is reversible and helps control metabolic processes. Researchers also study enzyme inhibitors as possible treatments for cancers that involve cell growth.
