Membrane Lipids

Nella membrana sono presenti tre tipi di lipidi: fosfogliceridi, sfingolipidi e steroli.
I fosfogliceridi, noti anche come glicerofosfolipidi, contengono una spina dorsale di glicerolo con un gruppo fosfato idrofilo e due catene aciliche grasse idrofobe, solitamente lunghe da 16 a 22 atomi di carbonio.
Alcuni dei fosfogliceridi più comuni presenti nella membrana cellulare sono la fosfatidilcolina, la fosfatidiletanolammina, la fosfatidilserina e il fosfatidilinositolo, formati dall'attaccamento di colina, etanolammina, serina e inositolo al gruppo fosfato.
Gli sfingolipidi sono lipidi che contengono una spina dorsale della sfingosina. La sfingosina è un amminoalcol con una lunga catena idrocarburica insatura. 
Lo sfingolipide più semplice è la ceramide, che si forma quando una molecola di acido grasso si attacca al gruppo amminico della sfingosina.
Il restante gruppo ossidrile della ceramide può formare esteri con altri gruppi chimici come la fosfocolina o i carboidrati, formando rispettivamente sfingomielina o glicolipidi.
Il terzo tipo di lipidi di membrana, gli steroli, come il colesterolo, non possono formare autonomamente un doppio strato lipidico. Si intercalano invece all'interno dei doppi strati fosfolipidici e fungono da tampone di fluidità per la membrana.

I lipidi sono una componente essenziale di tutte le membrane biologiche. Il contenuto medio di lipidi nelle membrane dei mammiferi è del 50%, anche se può essere del 20% nella membrana mitocondriale interna o dell'80% nella guaina mielinica presente attorno alle cellule nervose.

Fosfatidilcolina, fosfatidiletanolammina, fosfatidilserina e sfingomielina sono i fosfolipidi più comuni presenti nelle membrane dei mammiferi. A pH fisiologico la fosfatidilserina è carica negativamente, mentre le altre tre rimangono neutre. Nei lipidi di membrana il numero di atomi di carbonio presenti negli acidi grassi e il loro grado di saturazione possono variare. Gli acidi grassi possono essere completamente saturi se non presentano doppi legami, monoinsaturi se presentano un solo doppio legame o polinsaturi se hanno più doppi legami.

I glicolipidi comprendono una classe fisiologicamente importante di lipidi di membrana che sono sempre presenti sul lato esterno o extracellulare della membrana. Negli eucarioti si trovano sia nella membrana plasmatica che nelle membrane intracellulari. Il galattocerebroside, un glicolipide semplice, esiste abbondantemente nelle guaine mieliniche insieme alla sfingomielina. I gangliosidi, i glicolipidi più complessi, sono caricati negativamente a causa della presenza di acido sialico e possono costituire fino al 10% dei lipidi totali nelle membrane delle cellule nervose. I gangliosidi sono studiati per il loro ruolo nelle malattie virali e genetiche: il ganglioside GM1 agisce come recettore sulla superficie cellulare per la tossina del colera, mentre un eccessivo accumulo di gangliosidi GM2 si osserva nella malattia di Tay-Sachs, una malattia genetica.

Un'altra classe di lipidi di membrana sono gli steroli. Gli steroli vegetali costituiscono una componente significativa della membrana plasmatica delle piante e possono comprendere il 30-50% dei lipidi totali della membrana. Nelle membrane eucariotiche è presenti anche il colesterolo, che costituisce quasi il 50% dei lipidi totali. La struttura del colesterolo nelle membrane cellulari umane è leggermente diversa dall'ergosterolo, un comune costituente lipidico delle membrane fungine.

Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
