Asymmetric Lipid Bilayer

Il piccolo gruppo di testa della fosfatidiletanolammina gli consente di inserirsi nello strato interno della membrana curva. Al contrario, i grandi gruppi di testa di fosfatidilcolina e sfingomielina si inseriscono nella membrana curva esterna.

Le membrane biologiche presentano una distribuzione non uniforme di diversi tipi di lipidi negli strati interno ed esterno, risultando in membrane trasversali asimmetriche. Il trattamento della membrana eritrocitaria con l'enzima fosfolipasi ha confermato la natura asimmetrica del doppio strato lipidico. L'enzima idrolizza i lipidi in acidi grassi e gruppi idrofili. La fosfolipasi agisce solo sullo strato esterno della membrana, mentre lo strato interno rimane intatto. Il trattamento con fosfolipasi ha comportato una degradazione dell'80% della fosfatidilcolina totale di membrana (PC), mentre solo del 20% della fosfatidilserina (PS) e della fosfatidiletanolammina (PE). Il risultato sperimentale ha suggerito che nelle membrane degli eritrociti la maggior parte del PC è presente nello strato esterno, mentre il PS e il PE si trovano principalmente nello strato interno.

La distribuzione dei lipidi in ciascuno strato del doppio strato lipidico avviene in base alle loro esigenze funzionali, poiché lo strato interno e quello esterno di un doppio strato lipidico si trovano in ambienti diversi. Ad esempio, il PS caricato negativamente è necessario per il funzionamento degli enzimi intracellulari come la proteina chinasi C. Nello strato interno è presente anche il fosfatidilinositolo (PI); quando fosforilato, lega e localizza varie proteine citosoliche coinvolte nella segnalazione cellulare.

È noto che alcuni lipidi come gli sfingolipidi e il colesterolo formano microdomini nello strato esterno che assomigliano a piccole isole nel mare della membrana chiamate zattere lipidiche. Questi microdomini sono spesso associati ai glicoinositolfosfolipidi o alle proteine accoppiate a GPI, generando asimmetria della membrana laterale. Oltre ai lipidi, anche le proteine e i carboidrati contribuiscono all’asimmetria lipidica. Gli strati interno ed esterno sono associati a proteine diverse, mentre i carboidrati, sotto forma di glicoproteine e glicolipidi, sono sempre associati allo strato esterno, aumentando ulteriormente l'asimmetria della membrana.

Biological membranes have an uneven mix of lipids between the inner and outer layers. This membrane asymmetry gives each side a different chemical makeup and supports different cell functions.

Evidence for this uneven layout came from treating an erythrocyte membrane with phospholipase. Phospholipase is an enzyme that breaks lipids into fatty acids and hydrophilic groups. It acted only on the outer layer, while the inner layer stayed intact. The treatment degraded about 80% of total membrane phosphatidylcholine, or PC, but only about 20% of phosphatidylserine, or PS, and phosphatidylethanolamine, or PE.

These results show that most PC is in the outer layer of the erythrocyte membrane. In contrast, PS and PE are mainly in the inner layer. Lipid distribution follows functional needs because the two sides of the bilayer face different environments. Negatively charged PS helps intracellular enzymes such as protein kinase C work properly. Phosphatidylinositol, or PI, is also found in the inner layer. When PI is phosphorylated, it helps bind and localize cytosolic proteins involved in cell signaling.

Some lipids also cluster in the outer layer to form microdomains called lipid rafts. These rafts are made from sphingolipids and cholesterol and can look like small islands in the membrane sea. They are often linked with glycoinositolphospholipids, or GPI-coupled proteins, which adds to lateral membrane asymmetry. Proteins also differ between the inner and outer layers. Carbohydrates, in the form of glycoproteins and glycolipids, are found on the outer layer and further increase membrane asymmetry.
