Per determinare se due tratti sono stati ereditati separatamente o insieme, Mendel ha anche eseguito croci con piante di piselli che differivano in due tratti, come il colore del piselli e la forma del piselli. Per queste croci diibride, Mendel prima accoppiato piante che erano vero allevamento (cioè omozigoti) per diversi tratti delle stesse due caratteristiche. Mendel indusse poi l'autoimpollinazione nei diibridi F1. Delle sedici possibili combinazioni di allele parentali, nove producono prole con entrambi i tratti dominanti, piselli gialli e rotondi. Sei eventi di fecondazione conferiscono un tratto dominante, con tre prodotti gialli (dominanti), piselli rugosi e tre che creano piselli verdi e rotondi (dominanti). L'unica possibilità rimanente si traduce in piselli verdi e rugosi, i due fenotipi recessivi.

I geni su cromosomi separati e non omologhi sono assortiti in modo indipendente in gameti durante la meiosi.
