Cancer-critical genes are a group of genes that, upon mutation or alteration, contribute to the development of cancer.&#160;
These genes can be categorized into two major classes based on how the mutation affects gene activity - proto-oncogenes and tumor-suppressor genes.
Tumor-suppressor genes generally encode proteins that inhibit cell proliferation and trigger cell apoptosis &#8211; either directly or indirectly.
A loss-of-function mutation occurring in a single allele of the tumor-suppressor gene does not necessarily affect the gene&#8217;s overall function as the loss-of-function mutations act in a recessive manner.
Hence, both the alleles must lose their function to cause the uncontrolled proliferation of cells.
For example, the inactivation of the tumor suppressor gene, p53 has been identified in various human cancers, including leukemia and lymphoma.

Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype.
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.
Such genes that act as the drivers for cancer are termed cancer-critical genes and are categorized into two broad classes - proto-oncogenes and tumor suppressor genes. Tumor suppressor genes under normal conditions slow down cell division, repair errors in DNA, or control cell apoptosis. The loss-of-function mutations in tumor-suppressor genes make cells grow out of control and contribute to the development of cancer. Except for a few mutations in tumor-suppressor genes, which are inherited, most are acquired. Tumor suppressor genes like INK4, p53, and PTEN are very commonly mutated in many cancer types.
The major difference between proto-oncogenes and tumor suppressor genes is that proto-oncogenes lead to cancer upon over-activation, while tumor suppressor genes cause cancer when they are inactivated.

Tumor suppressor genes help keep cell growth under control. They can slow cell division, repair DNA errors, or trigger apoptosis, which is programmed cell death. These jobs protect healthy cells and help maintain normal cell cycle progression.

When a tumor suppressor gene is disrupted, cells can lose this control. Loss-of-function mutations in these genes can let damaged cells keep dividing. Over time, that unchecked growth can contribute to tumor formation and cancer.

Most tumor suppressor gene mutations are acquired, not inherited. However, a few can be passed down from one generation to the next. Tumor suppressor genes such as INK4, p53, and PTEN are often mutated in many cancer types.

Tumor suppressor genes are one major class of cancer-critical genes. The other class is proto-oncogenes. Proto-oncogenes can drive cancer when they are over-activated, while tumor suppressor genes lead to cancer when they are inactivated.
