Le biomolecole come le proteine e gli acidi nucleici sono composti a bassa volatilità con alti pesi molecolari. La spettrometria di massa convenzionale a ionizzazione a impatto elettronico deve affrontare sfide con tali molecole, tra cui una grave frammentazione molecolare.
La ionizzazione elettrospray è un metodo alternativo di ionizzazione "morbida" che genera ioni molecolari non radicalici per la spettrometria di massa di tali biomolecole.
In questo metodo, il campione viene miscelato con un solvente polare come metanolo, metanolo-acqua o acetonitrile con o senza sali aggiunti.
La miscela viene spruzzata attraverso un capillare ad alta tensione in una camera a vuoto. La rapida evaporazione del solvente dallo spruzzo fine di goccioline cariche porta infine alla molecola del campione gassoso carica.
Questa molecola del campione carica viene rilevata nello spettro di massa come somma della massa molecolare del campione e delle masse degli ioni associati.
Ad esempio, se gli ioni sodio sono presenti nella soluzione ionica, lo spettro presenta tipicamente un picco M+23.

Le biomolecole di peso molecolare più elevato sono composti non volatili che possono decomporsi prima di ionizzarsi o vaporizzarsi durante l'analisi di massa con i metodi convenzionali di ionizzazione per impatto elettronico. Di conseguenza, la ionizzazione elettrospray (ESI) è il metodo preferito per vaporizzare e ionizzare le biomolecole, poiché ne evita la rapida frammentazione e consente la registrazione dei segnali di massa per l'intera biomolecola.

Queste specie cariche vengono rilevate nello spettro di massa a un valore m/z equivalente alla somma della massa molecolare della molecola dell'analita e del peso molecolare degli ioni del liquido ionico associato alla molecola dell'analita.

La posizione dei segnali di massa varia a seconda del tipo di cationi presenti nel liquido ionico. Ad esempio, la presenza di ioni sodio nel liquido ionico porta a un picco a (M+23) negli spettri di massa, mentre la presenza di un protone porta a un picco a (M+1).

Electrospray ionization, or ESI, is a mass spectrometry method used to detect biomolecules that are too large or nonvolatile for conventional electron impact ionization. These molecules may break down before they can ionize or evaporate. ESI helps prevent rapid fragmentation and lets scientists record a mass signal for the whole biomolecule.

ESI uses electrical energy to move ions from the liquid phase into the gas phase. The analyte biomolecule is mixed with an ionic liquid, which is a nonmolecular compound that acts as the ionization source. The solution then passes through a high-voltage capillary and forms a fine aerosol.

A voltage difference between the capillary and the entrance to the analysis chamber pulls the charged droplets toward the chamber. As the droplets travel, the ionic liquid evaporates. This process leaves behind a protonated or otherwise ionized analyte molecule.

The charged species are measured in the mass spectrum by their m/z value, which reflects the analyte’s molecular mass plus the mass of the ions from the ionic liquid attached to it. The peak position depends on which cations are present in the ionic liquid. For example, sodium ions produce an (M+23) peak, while a proton produces an (M+1) peak.
