The Nucleus

Nelle cellule eucariotiche, il nucleo è un organello legato alla membrana che ospita le informazioni genetiche della cellula, il DNA.
Tuttavia, l'organizzazione del DNA all'interno del nucleo dipende dalla fase del ciclo cellulare.
Ad esempio, quando la cellula non si divide, i complessi proteici del DNA, noti come fibre di cromatina, sono sospesi liberamente nel nucleoplasma, consentendo un facile accesso alla replicazione del DNA e agli enzimi di trascrizione.
Al contrario, quando la cellula sta per dividersi, le fibre di cromatina vengono compresse in cromosomi.
Il nucleolo è una regione distinta all'interno del nucleo specializzata per l'assemblaggio dei ribosomi, complessi RNA-proteina responsabili della traduzione proteica.
Il nucleo è circondato da una doppia barriera di membrana, l'involucro nucleare, che è rivestito da complessi di pori nucleari o NPC.
Queste NPC controllano lo scambio di molecole tra il nucleo e il citoplasma.
Mentre la maggior parte delle cellule del corpo ha un nucleo, alcune, come i globuli rossi, sono anucleate, cioè mancano di un nucleo. Altri, invece, come le cellule muscolari scheletriche, sono multinucleati a causa di esigenze strutturali e funzionali.

Il nucleo è un organello legato alla membrana che funge da centro di controllo in una cellula eucariotica. Contiene DNA cromosomico, che controlla l'espressione genica e regola con precisione la produzione di proteine all'interno della cellula. Al contrario, il DNA all’interno dei mitocondri e dei cloroplasti svolge solo funzioni specifiche di quegli organelli.

Disposizione del DNA all'interno del nucleo

La regolazione dell'espressione genica all'interno del nucleo dipende da molti fattori, inclusa la struttura del DNA. Il DNA nel nucleo è avvolto attorno a proteine chiamate istoni, creando un complesso DNA-proteina chiamato cromatina. Quando le cellule non si dividono, cioè durante l'interfase, la cromatina è organizzata in modo diffuso. Ciò consente un facile accesso al DNA durante la trascrizione. Quando la cellula eucariotica è sul punto di dividersi, la cromatina si condensa strettamente in cromosomi lineari distinti, che possono essere facilmente segregati nelle cellule figlie.

Nucleoli

Il nucleolo è un organello privo di membrana che si forma attorno ai loci cromosomici dell'RNA ribosomiale all'interno del nucleo. Pertanto, i nucleoli sono il sito per la sintesi dell'rRNA e l'assemblaggio delle subunità ribosomiali. Queste subunità vengono successivamente esportate dal nucleo per essere assemblate in ribosomi completamente funzionali.

Ulteriori strutture subnucleari

Il nucleo contiene anche molti altri corpi nucleari, come i corpi di Cajal, speckles e paraspeckles. Questi corpi nucleari aiutano a compartimentalizzare spazialmente l'ambiente nucleare e creare siti distinti per specifiche reazioni biologiche. The nucleus is a membrane-bound organelle that stores chromosomal DNA and helps control what proteins a eukaryotic cell makes. It acts as the cell’s control center. DNA in mitochondria and chloroplasts is different because it mainly supports functions specific to those organelles.

Inside the nucleus, DNA is wrapped around proteins called histones. This DNA-protein complex is called chromatin. During interphase, when the cell is not dividing, chromatin stays loosely arranged so the DNA is easy to access for transcription. As a eukaryotic cell prepares to divide, the chromatin condenses into distinct, linear chromosomes that can be separated into daughter cells.

The nucleus also contains the nucleolus, which is a membrane-less organelle. It forms around chromosomal regions that contain ribosomal RNA, or rRNA. The nucleolus is the site of rRNA synthesis and the assembly of ribosomal subunits. These subunits later leave the nucleus and join together to form functional ribosomes.

Other nuclear bodies are present as well. Cajal bodies, speckles, and paraspeckles help organize the nuclear space into regions for specific biological reactions. Their membrane-less structure also allows materials to move more easily with the nucleoplasm, the fluid inside the nucleus.
