Gene Evolution - Fast or Slow?

Il modo in cui le mutazioni influenzano la sopravvivenza di una cellula dipende fortemente dalla posizione del cambiamento genetico. In regioni del genoma che non codificano per geni o regioni regolatrici le mutazioni possono avere scarso effetto, e lo sviluppo della cellula possa spesso continuare normalmente. Sebbene alcune di queste regioni contengano sequenze regolatrici fondamentali, la stragrande maggioranza di questo DNA non svolge alcuna funzione nota. Tipicamente, queste regioni del genoma sono quelle in cui si osserva il cambiamento più rapido, in termini evolutivi, perché tipicamente su queste regioni agisce una pressione selettiva minima o nulla per preservarne le sequenze.

Al contrario, le regioni che codificano per una proteina potrebbero essere sottoposte a un'elevata pressione selettiva, poiché qualsiasi cambiamento nella loro sequenza potrebbe risultare in una proteina meno capace di svolgere la sua funzione in modo ottimale. Tuttavia, occasionalmente, una mutazione in una di queste regioni si tradurrà in un risultato benefico che contribuisce alla forma fisica generale dell’organismo, tali mutazioni spesso persistono e possono persino diventare fisse nelle popolazioni. Quando si confronta la frequenza di questi eventi di mutazione con i cambiamenti relativamente regolari osservati nelle sequenze non codificanti, questo è estremamente raro e quindi in generale le regioni codificanti sono considerate come ad evoluzione lenta.

È anche vero che esiste una quantità misurabile di variazione nei livelli di conservazione delle sequenze all'interno delle sequenze codificanti, e questo si osserva in tutti gli organismi. Prendiamo, ad esempio, una proteina recettore. Tali proteine hanno tipicamente regioni diverse che possono svolgere funzioni come il legame del ligando, la segnalazione intracellulare o l'integrazione della membrana. In questo caso, una mutazione nella regione coinvolta nel legame del ligando può produrre una proteina meno efficiente nel legare il ligando. Pertanto, la pressione selettiva sarebbe probabilmente elevata sui particolari nucleotidi che codificano per questa parte della proteina. Tuttavia, nella sezione della proteina che attraversa la membrana, si potrebbe osservare un effetto minore se si verifica una sostituzione di amminoacidi, e quindi livelli più bassi di pressione selettiva. In queste condizioni, potremmo vedere che due regioni dello stesso gene codificante una proteina potrebbero avere velocità di evoluzione diverse.

Sequenziamento di Geni o Regioni Genomiche per Costruire Filogenesi

Questa variazione nella velocità dell'evoluzione del genoma in diverse regioni può essere studiata per rispondere a domande sulle relazioni evolutive. I geni e le regioni genetiche possono essere selezionati e sequenziati su gruppi di individui per rispondere a domande ristrette come "queste popolazioni sono potenzialmente specie diverse?" o ampie come "come si collocano questi phyla nell'albero della vita?". Nel primo caso, la selezione di un gene che presenta una regione relativamente poco conservata aiuterebbe a identificare le differenze a livello di popolazione. Al contrario, per rispondere a domande su gruppi diversi come i phyla, una regione genetica altamente conservata può fornire un’omologia sufficiente per produrre una filogenesi di tali gruppi. Le regioni comunemente utilizzate per analisi filogenetiche molecolari come queste includono geni dell'rRNA ribosomiale (come rRNA 16s, rRNA 18s o rRNA 28s) o regioni genomiche note come ITS (Distanziatori Trascritti Interni, I o II) che si trovano tra i geni della subunità dell'rRNA ribosomiale.

Gene evolution can happen at different rates, and natural selection helps shape those changes. Some genes change slowly because important DNA changes are removed over time. Other genes can change more quickly when different mutations are kept in a population.

The rate of evolution depends on how a gene is used and how strongly it is affected by selection. Genes with essential jobs often stay more similar across species. Genes under weaker pressure may show more variation and may evolve faster.

Comparing gene sequences helps scientists see these patterns over time. These comparisons can show which regions are conserved and which regions have changed more rapidly. That information helps explain how genes evolve in different lineages.
