Le caderine sono una superfamiglia di molecole di adesione cellulare calcio-dipendenti o CAM .
Tutte le caderine hanno una struttura di base che comprende un piccolo dominio citosolico C-terminale, un singolo dominio transmembrana e più domini extracellulari di caderina o EC.
Questi domini EC sono collegati tra loro da regioni cerniera flessibili, ciascuna con tre tasche leganti il calcio.
Quando gli ioni calcio si legano, la caderina diventa rigida, consentendo il cis e il trans-legame con altre caderine.
Nel cis-binding, le caderine sulla stessa cellula possono legarsi l'una all'altra formando cluster, mentre l'interazione dei domini EC delle caderine sulle cellule adiacenti provoca il trans-legame.
Il dominio EC N-terminale, il più lontano dalla membrana, chiamato EC1, ha una struttura a manopola e tasca. Durante il trans-binding, la manopola di un EC1 si inserisce nella tasca di un altro EC1 sulla cella adiacente.

Structure of Cadherins

Le caderine furono una delle prime molecole di adesione cellulare scoperte; il termine “caderine” si basa sulle loro proprietà di adesione calcio-dipendenti. Le prime caderine scoperte sulle cellule epiteliali, neuronali e placentari furono chiamate rispettivamente E-caderina, P-caderina e N-caderina. Queste caderine classiche condividono sequenza e somiglianze strutturali. Altre caderine, comprese quelle coinvolte nella segnalazione cellulare, sono raggruppate in caderine non classiche. Questa diversità di caderine deriva da più geni della caderina nel genoma e dallo splicing alternativo dell'RNA durante la trascrizione.

Caderine nelle giunzioni cellulari

Le caderine sono proteine transmembrana con molteplici domini leganti extracellulari. Il loro dominio citosolico lega le proteine linker che si collegano al citoscheletro. Le caderine si raggruppano sulla membrana plasmatica legandosi in cis, formando grandi complessi multiproteici chiamati giunzioni cellulari. Mentre le caderine classiche come la E-caderina formano giunzioni aderenti, le caderine non classiche come la desmogleina e la desmocollina formano giunzioni desmosomiali. Queste giunzioni cellulari funzionano per tenere insieme le cellule e fornire resistenza meccanica ai tessuti, come l'epitelio.

Significato clinico delle caderine

Le caderine svolgono un ruolo essenziale nell'adesione cellulare, nella formazione dei tessuti e nella comunicazione intercellulare. Di conseguenza, qualsiasi cambiamento nella loro struttura, espressione e stabilità può manifestarsi in condizioni cliniche. Quando le cellule perdono il contatto con le cellule vicine, possono liberarsi dal tessuto e invadere altri tessuti. La perdita di adesione cellulare dovuta a mutazioni nei geni della caderina è associata alla malignità dei tumori, come quelli osservati nelle cellule maligne del seno. Gli agenti patogeni utilizzano anche le caderine come recettori per attaccarsi e invadere la cellula ospite. Ad esempio, la caderina CDHR3 è altamente espressa nell’epitelio polmonare e viene utilizzata come recettore dal rinovirus di classe C (RV-C) che causa il comune raffreddore.

Cadherins are cell adhesion proteins that help cells stick together and build tissues. Their name comes from their calcium-dependent binding. Early examples were named E-cadherin, P-cadherin, and N-cadherin after the epithelial, placental, and neuronal cells where they were first found.

Classical cadherins share similar sequences and structures. Other cadherins, including those involved in cell signaling, are grouped as non-classical cadherins. This diversity comes from multiple cadherin genes in the genome and from alternative RNA splicing during transcription.

Cadherins are transmembrane proteins. They have several extracellular binding domains and a cytosolic domain inside the cell. The cytosolic region binds linker proteins, which connect cadherins to the cytoskeleton. Cadherins also cluster on the plasma membrane through cis-binding, which helps form large protein complexes called cell junctions.

Classical cadherins such as E-cadherin form adherens junctions. Non-classical cadherins such as desmoglein and desmocollin form desmosomal junctions. These junctions hold cells together and give mechanical strength to tissues such as epithelium.

Cadherins are important for cell adhesion, tissue formation, and communication between cells. Changes in their structure, expression, or stability can lead to disease. When cells lose contact with neighboring cells, they may break free from the tissue and invade other tissues. Mutations in cadherin genes are linked to tumor malignancy, including in breast cancer cells. Some pathogens also use cadherins as receptors to attach to and enter host cells. One example is CDHR3, which is highly expressed in lung epithelium and is used by class-C rhinovirus, or RV-C, the virus that causes the common cold.
