La giunzione aderente è una giunzione di ancoraggio che tiene insieme le cellule in tessuti come l'epitelio.
Le giunzioni aderenti sono grandi complessi multiproteici che comprendono tre classi proteiche principali: le caderine adesive, il citoscheletro di actina di ancoraggio e le catenine linker.
L'assemblaggio della giunzione aderente inizia con la polimerizzazione dei monomeri di actina vicino alla membrana cellulare per formare sporgenze chiamate lamellipodi.
Quando i lamellipodi delle cellule vicine sono abbastanza vicini, le caderine sulla loro superficie interagiscono e innescano la localizzazione di più caderine, formando grappoli.
Questi cluster sono stabilizzati da catenine beta, alfa e p120, che aiutano a reclutare più complessi caderina-catenina nel sito di crescita.
Man mano che la zona di contatto tra le due cellule si espande, le alfa-catenine spostano la polimerizzazione dell'actina da ramificata a non ramificata e le dispongono parallelamente alla membrana cellulare.
Questi filamenti di actina si associano quindi alla miosina per formare fasci contrattili, formando così una giunzione aderente matura.
La giunzione collega il citoscheletro di una cellula con un'altra, tenuto insieme tramite le interazioni extracellulari delle molecole di caderina.

Adherens Junctions

Forti punti di contatto tra cellule adiacenti le ancorano tra loro, formando i tessuti. Tali giunzioni di ancoraggio sono di due tipi: giunzioni aderenti e desmosomi. Le giunzioni aderenti sono abbondanti nei tessuti come l'epitelio e l'endotelio, formando una zona continua di adesione chiamata cintura di adesione. In altri tessuti, come il muscolo cardiaco, appaiono come grappoli, che collegano le cellule per produrre una contrazione coordinata del muscolo cardiaco.

Le giunzioni aderenti sono dinamiche

Le cellule endoteliali che rivestono i vasi sanguigni sono strettamente collegate, formando una barriera che impedisce la fuga di altre cellule e molecole dal flusso sanguigno nel tessuto circostante. Tuttavia, durante una risposta immunitaria, i leucociti del flusso sanguigno devono attraversare questa barriera e raggiungere il tessuto bersaglio. Le molecole infiammatorie come l'istamina e le proteine di segnalazione come il fattore di crescita dell'endotelio vascolare (VGEF) innescano processi a valle, come la fosforilazione e l'internalizzazione delle caderine presenti sulla superficie delle cellule endoteliali. Ciò porta al disassemblaggio delle giunzioni aderenti e aumenta la permeabilità endoteliale creando spazi che consentono il passaggio delle cellule immunitarie e di altre molecole. Il mantenimento e la rottura delle giunzioni aderenti sono quindi regolati dinamicamente in risposta a stimoli specifici.

Giunzioni aderenti e infezione

Agenti infettivi come l'Helicobacter pylori possono entrare nel tessuto ospite distruggendo le giunzioni aderenti e muovendosi attraverso gli spazi intercellulari. Questi batteri producono proteasi che fendono il dominio extracellulare delle caderine, causando così il disassemblaggio della giunzione. Le caderine della superficie cellulare vengono anche dirottate come recettori da batteri come lo Streptococcus pneumoniae per aderire all'epitelio respiratorio superiore.

Tuttavia, il dirottamento più impressionante è forse quello dimostrato dalla Listeria monocytogenes. Questo batterio esprime proteine della parete cellulare che attaccano le E-caderine e innescano il reclutamento delle catenine ɑ, β e p120 nel sito. Questo reclutamento avvia la polimerizzazione dinamica dell'actina, formando sporgenze della membrana che inghiottiscono i batteri in un processo chiamato fagocitosi indotta. Il batterio riesce così ad entrare in una cellula non fagocitica e prolifera all'interno dell'ospite.

Adherens junctions are strong cell-to-cell anchors that help tissues stay connected. These anchoring junctions work with desmosomes, another type of junction that holds adjacent cells together. In many epithelial and endothelial tissues, adherens junctions form a continuous adhesion belt. In heart muscle, they often appear as clusters that help cells work together for coordinated contraction.

Adherens junctions are also dynamic, especially in the cells that line blood vessels. These endothelial cells form a barrier that normally keeps cells and molecules inside the bloodstream. During an immune response, leukocytes, or white blood cells, must cross this barrier to reach damaged tissue. Inflammatory molecules such as histamines and signaling proteins such as vascular endothelial growth factor, or VEGF, can trigger phosphorylation and internalization of cadherins on the endothelial cell surface. Cadherins are adhesion proteins that help cells stick together. When this happens, adherens junctions break down, permeability increases, and gaps open for immune cells and other molecules to pass through.

Some infections also take advantage of adherens junctions. Helicobacter pylori can disrupt these junctions by producing proteases that cut the extracellular part of cadherins. This breaks the junctions and helps the bacteria move through the spaces between cells. Other bacteria, such as Streptococcus pneumoniae, use cadherins as receptors to attach to the upper respiratory epithelium. Listeria monocytogenes uses a more complex strategy. It binds E-cadherins and recruits alpha-, beta-, and p120-catenins, which starts actin polymerization. This creates membrane protrusions that engulf the bacteria in induced phagocytosis, allowing it to enter a nonphagocytic cell and multiply inside the host.
