Cohesins

Durante la fase S del ciclo cellulare negli eucarioti, la duplicazione cromosomica produce due copie identiche di ciascun cromosoma chiamate cromatidi fratelli.
I cromatidi fratelli formati sono tenuti insieme da un insieme di complessi proteici chiamati coesine. Gli anelli di coesina esistono come morsetti attorno ai cromatidi fratelli in più punti lungo la loro lunghezza, impedendo loro di allontanarsi.
I complessi di coesina contengono quattro subunità: Smc1, Smc3, Scc1 e Scc3. Smc1 e Smc3 sono proteine a spirale con un dominio cerniera a un'estremità e un dominio testa ATPasi all'altra estremità. I domini cerniera di Smc1 e Smc3 si legano direttamente l'uno all'altro, mentre una subunità Scc1 collegata a una subunità Scc3 collega i domini di testa di Smc1 e Smc3, formando una struttura ad anello.
Il dominio cerniera della struttura dell'anello di coesina può essere attivato per aprirsi e chiudersi, facilitando il caricamento della coesina sui cromosomi.
Man mano che le cellule progrediscono attraverso la profase mitotica, avviene la risoluzione dei cromatidi fratelli, che comporta la dissociazione degli anelli di coesina lungo i bracci cromosomici preservando quelli legati alla regione del centromero. La rimozione differenziale delle coesine fa sì che i cromatidi fratelli si separino parzialmente lungo i loro bracci, pur rimanendo legati al loro centromero.
La coesione cromatidica al centromero facilita il bi-orientamento dei cromosomi sul fuso mitotico durante la metafase, garantendo il corretto attaccamento dei microtubuli ai cinetocori dei cromatidi fratelli.
All'inizio dell'anafase, un enzima proteasi, la separasi, scinde la subunità Scc1, portando alla dissociazione della coesina dal cromosoma.
La dissociazione della coesina permette la segregazione dei cromatidi fratelli durante l'anafase, dove vengono separati da fusi mitotici ai poli opposti della cellula, portando infine alla divisione cellulare e alla formazione di due cellule figlie.

coesine

I complessi proteici di coesione sono una colla molecolare che tiene insieme due cromatidi fratelli. Svolgono un ruolo importante sia nella mitosi che nella meiosi. Nella mitosi tutti i complessi di coesione presenti sui cromosomi vengono rimossi prima dell'inizio della fase anafase.

Complessi di coesione nella divisione meiotica

La meiosi coinvolge due cicli distinti di segregazione cromosomica e divisioni cellulari – Meiosi I seguita da Meiosi II – producendo quattro cellule figlie. La meiosi I include la separazione dei cromosomi omologhi, mentre la meiosi-II comporta la separazione dei cromatidi fratelli.

Il complesso di coesione della Meiosi I è costituito da quattro subunitàーSmc1, Smc3, Rec8 (che sostituisce Scc1 dal complesso di coesione mitotico) e Scc3 ー che formano una struttura ad anello.

Durante la Meiosi I, interi cromosomi si segregano verso i poli opposti poiché la rimozione della coesione avviene solo dai bracci cromosomici. La coesione è ancora mantenuta nella regione del centromero, consentendo ai cromatidi fratelli di rimanere collegati. Durante la transizione dalla metafase I all'anafase I, la rimozione differenziale della coesione è facilitata dalla scissione mediata dalla separazione della subunità Rec8 delle coesine lungo i bracci cromosomici. Il centromerico Rec8 è protetto dalla scissione mediante associazione con una proteina protettrice Shugoshin (Sgo1).

Coesinopatie

Le coesine contribuiscono al mantenimento della stabilità genomica. Le mutazioni nei geni che codificano per le subunità di coesione o i cofattori di coesione possono portare a malattie chiamate coesinopatie. La sindrome di Cornelia de Lange (CdLS) e la sindrome di Roberts sono le due coesinopatie meglio descritte. La CdLS è un disturbo dello sviluppo neurologico che causa ritardo mentale, dismorfismi facciali, anomalie degli arti superiori e ritardo della crescita. La sindrome di Roberts provoca anomalie cranio-facciali, riduzione degli arti e ritardo della crescita nei pazienti affetti.
