Forces Acting on Chromosomes

Durante la mitosi, i cromosomi biorientati oscillano per stabilizzare i loro attacchi al fuso e successivamente si assemblano lungo la piastra di metafase all'equatore del fuso mitotico.
Forze multiple agiscono sui cromosomi dopo che sono stati attaccati al fuso mitotico.
Una grande forza verso il polo esercitata lungo il cinetocoro-microtubulo, come risultato della depolimerizzazione dell'estremità positiva, attira il cinetocoro e il suo cromosoma associato verso il polo del fuso. Al cinetocore, i complessi proteici Ndc80 collegano il cinetocore al microtubulo attraverso molteplici attacchi a bassa affinità lungo i lati dei microtubuli.

Una terza forza, la forza di espulsione polare o vento polare, generata dalla chinesina-4 e dalle proteine motrici 10 spinge i cromosomi lontano dai poli del fuso. I bracci cromosomici di Kinesin-4 e 10 si collegano con microtubuli interpolari. Queste proteine motorie dirette all'estremità positiva spostano il cromosoma verso l'equatore del fuso.
Un'interazione bilanciata di queste molteplici forze opposte consente ai cromosomi biorientati di allinearsi con precisione lungo la piastra di metafase, in preparazione alla segregazione cromosomica.

Durante la mitosi, i movimenti cromosomici avvengono attraverso l'interazione di molteplici forze a livello di piconewton. Nella prometafase, queste forze aiutano nell'assemblaggio o nella riunione dei cromosomi sul piano equatoriale, portando infine al loro allineamento sulla placca metafase. Le forze che agiscono sui cromosomi dipendono dallo spazio e dal tempo; pertanto, variano con la posizione dei cromosomi man mano che la cellula progredisce attraverso la mitosi.

I microtubuli e le proteine motrici esercitano due tipi di forze sui cromosomi: verso i poli e anti-poli, note anche come forze di espulsione polare. La depolimerizzazione dei microtubuli del cinetocore genera la forza verso il polo e tira il cromosoma verso il polo del fuso. Al contrario, la polimerizzazione del cinetocore-microtubulo porta a forze di espulsione polare, che spingono il cromosoma verso l’equatore della cellula. Anche le proteine motorie dirette all’estremità positiva dei microtubuli, come le cromochinesine e la chinesina-7, producono forze di espulsione polare spingendo i cromosomi verso l’equatore della cellula.

L'azione simultanea ma disuguale delle forze verso i poli e delle forze di espulsione polare provoca l'oscillazione dei cromosomi durante la prometafase; tuttavia, durante la metafase, i cromatidi fratelli biorientati sperimentano forze uguali ma opposte. Ciò crea una tensione sufficiente per silenziare il percorso del punto di controllo dell'assemblaggio del fuso e consente alle cellule di passare all'anafase. Nell'anafase, le forze verso i poli agiscono sui cromatidi fratelli, determinandone la segregazione nelle cellule figlie.

Oltre alle forze di cui sopra, i cromosomi sono soggetti anche a forze di coesione e di risoluzione.
